JAK inhibitors

Drug class that blocks Janus kinases, which are involved in cytokine signalling. It includes baricitinib (JAK1/JAK2) and ritlecitinib (JAK3/TEC), which are not mechanistically interchangeable. The EMA and the FDA have attached safety warnings to this class regarding serious infections, thromboembolism, major cardiovascular events and malignancies.
